IDENTIFICATION OF THE POPULATION GENETIC STRUCTURE OF PATHOGENIC ORGANISMS OF TURKEY DISEASES

It is planned to carry out a research work in the Genetic Selection Centre “North
Caucasus Zonal Experimental Station of Poultry” which is aimed at the prevention and control of
infectious diseases of birds, causing huge economic damage to the industry. The creation of
domestic competitive lines and crosses of birds requires a reliable system of livestock protection,
which can be based on the use of modern molecular genetics techniques, including genotyping
and PCR tests of antibiotic resistance of pathogenic microorganisms. The research is based on
the use of the method of genotyping of pathogenic microorganisms, which uses restriction
endonucleases with simultaneous labeling of individual DNA fragments, which are then
visualized on a nylon filter. During the research, cultures of bacterial isolates were obtained in
elective, selective and differential diagnostic media in the Department of Microbiology of AllRussian Research Veterinary Institute of Poultry Science after sampling and biomaterials from
turkeys of different age groups in the Genetic Selection Centre “North Caucasus Zonal
Experimental Station of Poultry”. The species of bacteria were also determined with the help of
cultural and molecular genetics techniques (standard PCR with primers to 16S DNA of bacteria);
the comparison of genotyping data by two different methods was carried out; an in silico search
of restriction endonucleases for genotyping by the DREAM technique was carried out; E. coli
bacteria were genotyped using combinations of restriction endonucleases XhoI / BsuRI and XbaI
/ PstI, with the identification of bacterial genotypes.
